It would be easy to simply describe League of Legends as the de facto sequel to Defense of the Ancients (DOTA), the wildly successful Warcraft 3 mod that rivaled the game it was based on for complexity and popularity. It would also be wrong. Despite the involvement of some of that game's originators, League builds substantially on that game while adding a multitude of completely original features.
It's probably telling that the most hardcore of the hardcore DOTA players seem to show preference for Heroes of Newerth, which cleaves more closely to its spiritual predecessor. League, on the other hand, has more broad appeal. It's still an intimidatingly complex game, with dozens upon dozens of characters, items, masteries, and runes to figure out and work into the most advantageous combination to match individual playstyles. But on the whole they're so well balanced that there's actually room for dicking around a bit without ending up completely gimped. Contrast with HON and DOTA, which have very narrow tolerances for success and a community of players who will often volunteer some surprising information about your mother should you have the audacity to be the first one to fall in a match. If you don't like those guys, well, they're probably playing something else.
The initial impression, especially for non-veteran players, might be something like "Who are all these guys? What are all these things? What does everything do? For the love of crap, my head can't hold it all!" But between a convenient auto-matching system that throws together players of similar skill, as well as little touches like suggested characters for new players and suggested items at the weapon shop, make entry relatively painless. Meanwhile, if you did happen to be good at DOTA (distinct from "just having played DOTA"), well, you will most likely be pretty good at this right out of the gate.

The biggest change to the formula, however, is the persistence of your player as a "Summoner," not to be confused with the 30-odd in-game characters, which get a clean slate at the start of every match. You, yourself, have a skill level (raised through XP gained from matches), and acquire points to be spent on three trees of masteries. Some of these are traps for the uninformed -- like "Greed," which is simply useless. Most, however, are fairly well balanced and add to the individuality of the players. Runes and spells are similarly acquired through play, and can be swapped into different loadouts for each match. It seems a lot to deal with at first, but they're all gained at a pace that prevents things from becoming overwhelming, and provide something to work toward beyond simple ranking.
Which is good, because ranked matches aren't implemented yet. In fact, you'll be seeing a lot of grayed-out options in the menus. At present there's only one map and game mode available, and the game's store won't be online until the November 17. Though that last one is hard to complain about, given that the characters one would be gaining access to through microtransactions are now all available to everyone until that opening date. Afterward, there will be eight characters available to all players -- on a rotating basis.
Which is all pretty damn spiffy for a free game; and the boxed edition, which includes access to two-thirds of the game's characters and a $10 card for the game's store, is a pretty cost-effective buy if you find yourself hooked. Which is likely, given that League actually pulls off the trick of making itself much more accessible than its predecessors and rivals, without ever coming off as dumbed down.
Really, this thing isn't providing me with more than the usual gripes about multiplayer strategy/defense games. You know, stuff like how when a player drops from your side it means all but sure defeat, and how surrenders are far more common and less satisfying than a game played through to its ultimate conclusion. Oh, and I still completely suck, but that's my own fault.
